Process analytical methods for monitoring of the film coating on pharmaceutical pellets and tablets using NIR spectrometry and image analysis

Abstract
Pharmaceutical tablets and pellets are often covered with a coating. There is a need for methods that enable monitoring of the growth of such films during processing. Such analyses can be performed using process analytical chemical methods based on near infrared (NIR) diffuse reflectance spectrometry. It provides with analyses that take only ca. 8 seconds in total analysis time, including physical sampling. The use of chemometrics is the key to handle the complexity of the NIR spectra. Pharmaceutical tablets and pellets are often covered with a coating. There is a need for methods that enable monitoring of the growth of such films during processing. Such analyses can be performed using process analytical chemical methods based on near infrared (NIR) diffuse reflectance spectrometry. It provides with analyses that take only ca. 8 seconds in total analysis time, including physical sampling. The use of chemometrics is the key to handle the complexity of the NIR spectra. The chemometric methods of principal component analysis (PCA) and partial least squares (PLS) were used to study and to calibrate NIR-spectra to the amount of coating. An at-line method using NIR-spectra for determination of the amount of coating of film applied on pharmaceutical tablets was developed. Limitations for quantitation of the film were determined by the 'information depth', ca 0.1 - 0.2 mm.



Novel tools to determine the coating thickness and its variations on pharmaceutical pellets were based on image analysis and NIR diffuse reflectance spectrometry. Data from the images were used to predict variations in the release rate of the active substance due to geometrical variations. The image analysis was further used as a reference method for thickness measurements. Together with a theoretical film thickness growth model with physical relevance to underlying process mechanisms, an in-line process analytical chemical method based on NIR was enabled for analysis of the coating thickness on pharmaceutical pellets (RMSEP = 2.2 µm, range tested 0 - 50 µm). Indeed, such tools have not been combined previously, and through use of a multivariate batch calibration, the thickness values delivered from the NIR spectrometer became less disturbed by commonly varied process conditions. Additionally, variations in the coating thickness could be indicated by NIR . The spectral artefacts encountered for samples moving during acquisition of a scan were characterised in a separate study using multivariate methods. These artefacts could be detected by PCA. (Less)

Eftersom de uppmätta egenskaperna ofta används för att styra processandet mot bästa möjliga betingelser är det av generellt intresse att mätningen går snabbt. Men det är inte nog med att det går snabbt, man ska ju också kunna lita på siffrorna som kommer från mätningen. För att haltmätningar ska kunna fungera måste man därför kunna skilja på de olika ämnena.



Nära infrarödspektrometri, ofta förkortat till NIR, är en av många metoder för att snabbt kunna mäta halter i en kemisk process är . Med denna metod tar det bara ett par sekunder mellan varje uppmätt värde, vilket i kemiska sammanhang anses som mycket snabbt. Det mänskliga ögat kan inte se "färgerna" i NIR-området, utan som bekant enbart de färger som syns i regnbågen, från violett till rött. NIR ligger strax utanför det röda och man kan faktiskt uppfatta denna strålning trots att man inte kan se den genom att placera handen över en het spisplatta! Vissa speciella material kan registrera dessa för människan osynliga färger och även deras intensitet. På detta vis kan den osynliga färgen och dess intensitet registreras i form av en kurva där "färg" och intensitet kan avläsas. Om de olika ämnenas kurvor bara har olika form kan man använda dem för att bestämma ämnenas halter. Det är ofta en fördel om en så stor del som möjligt av denna kurva, detta fingeravtryck av provet, kan användas. Eftersom de olika ämnenas NIR-kurvor oftast har små nyans-skillnader utnyttjas s.k. multivariata metoder. Med dessa används hela kurvan och vikter beräknas för varje uppmätt intensitet längs med kurvan. Vikterna kombineras därefter med de uppmätta intensiteterna och summeras slutligen för att ge ett värde på exempelvis halten av ett ämne. Teorin för multivariata metoder upplevs ofta som komplicerad eftersom man brukar förklara dem med hjälp av mång-dimensionella rum vilka ju är svåra att begripa. Men genom att överföra vår uppfattning om hur det ligger till med till exempel avstånd i ett tredimensionellt rum, kan man med matematikens hjälp utan problem arbeta med många dimensioner, t ex 1000 eller fler. Men icke att förglömma, i slutänden är nyttan med multivariata metoder att bestämma vikter längs med den uppmätta kurvan.



I denna doktorsavhandling har uppgiften varit att ta fram process-analytiska metoder för att mäta filmtjocklekar på farmaceutiska material. Filmen kan användas som en skyddande beläggning, men anledningarna till att man över huvudtaget lägger på en film kan vara flera. Till exempel, genom att ha den aktiva substansen (det aktiva läkemedlet) i form av små kulor kan upplös-ningen i tarmen av det aktiva ämnet kontrolleras om en beläggning, en bromsande film läggs runt dessa kulor. Om filmen är alltför tunn blir motståndet för litet och det aktiva ämnet frigörs alltför snabbt och vice versa. Om tjockleken kan kontrol-leras innebär detta i slutänden att man har goda möjligheter att tillverka kapslar eller tabletter som har egenskapen att man bara behöver ta "en om dagen" istället för flera. Från varje tablett pyr det då långsamt ut av det aktiva ämnet. Genom att mäta med NIR på farmaceutiska processer där materialen beläggs med filmer kunde en processanalytisk metod utvecklas för snabb mätning av den aktuella filmtjockleken. För att kunna bestämma de olika vikterna längs med NIR-kurvan så att värden på filmtjockleken kunde levereras, användes multivariat kalibrering. För att denna kalibrering skulle resultera i att värdena från NIR-mätningen härrörde från enbart den pålagda filmen användes två viktiga delar. För det första behövdes en referensmetod, vilken bestod i att mäta filmtjockleken på fotografier med en dators hjälp. För det varierades systematiskt faktorer som kan påverka NIR-kurvans utseende. Därmed kunde vikterna längs med NIR-kurvan relteras till filmtjocklecken och samtidigt justerades de för att borstse från störande faktorer. Intensiteten längs med kurvan viktades alltså enligt ovan för att slutligen ge värden som var starkt knutna till filmtjockleken. (Less)
