Chondrocyte-matrix interactions: with emphasis on the collagen binding integrin a10ß1

Abstract
The work in this thesis has been focused on chondrocyte integrins and their interaction with the matrix proteins chondroadherin (CHAD) and collagen type II (CII). Affinity purification of CHAD-binding proteins from chondrocyte lysate, followed by immunoprecipitation of EDTA-eluted material identified alpha2beta1 as a CHAD-binding integrin. The work in this thesis has been focused on chondrocyte integrins and their interaction with the matrix proteins chondroadherin (CHAD) and collagen type II (CII). It is also demonstrated that alpha10 exist as two splice variants. Both splice forms are present in alpha10 expressing tissues, although the shorter form predominated in cells from articular cartilage and aortic valve as judged by PCR experiments. The longer form was upregulated when chondrocytes dedifferentiated in monolayer culture. The exon/intron boundaries of the mouse alpha10 gene was characterized and the gene was mapped to mouse chromosome 3F2.2 and human chromosome 1q21. (Less)

Brosk är en specialiserad form av bindväv som bekläder alla ledytor i kroppen och dess specifika sammansättning förser leden med en i det närmaste friktionslös rörelseförmåga. Broskets stötdämpande förmåga skyddar det underliggande benet genom att fånga upp och fördela krafterna som bildas vid belastning av en led. Bindväven i brosk (extracellulär matrix) består av ett nätverk olösliga kollagenfibrer i vilket ett fåtal celler och en mängd lösliga proteiner och proteoglykaner är fångade.



Kollagenfibrerna fungerar som armering och står för draghållfastheten i vävnaden. De lösliga proteoglykanerna består av en proteindel med påkopplade kolhydratkedjor sk glykosaminoglykaner (GAG). Den dominerande proteoglykanen i brosk heter aggrecan. GAG-kedjorna är negativt laddade och drar till sig vatten. Isolerat aggrecan i vattenlösning sväller upp till fem gånger sin volym i brosk. Denna expandering och de repellerande negativa laddningarna bidrar till att utrymmet mellan kollagenfibrerna fylls ut fullständigt, och därigenom förses brosket med dess stötdämpande funktion.



Cellerna i bindväv, vilka i brosk kallas chondrocyter, producerar och organiserar extracellulär matrix. De bryter även ner produkter som inte längre behövs. Det pågår ett ständigt samspel mellan uppbyggnad och nedbrytning av molekyler. Denna process är mycket noggrant kontrollerad och rubbningar leder till att broskets speciella egenskaper förändras. Detta kan i sin tur leda till att brosket försvagas och förstörs.



Integriner tillhör en familj av protein som finns i cellens membran. De är uppbyggda av två separata delar en alpha- och en beta- kedja som samarbetar med varandra. En del av integrinet finns på utsidan av cellen och förankrar cellen till bindvävs-proteiner. Den mosatta delen av integrinet finns inuti cellen och är där i kontakt med cellens signaleringsmaskineri. Integriner har därmed förmågan att förmedla information både utifrån bindväven till cellens insida samt inifrån cellen och ut till bindväven. Denna tvåvägskommunikation är av stor vikt både för nybildning och underhåll av vävnaden. Varje celltyp har sin specifika uppsättning av integriner som gör att de kan fästa och förmedla sin information i den vävnad där de hör hemma. Målet med denna avhandling har varit att öka förståelsen för hur broskceller kommunicerar med omgivningen med hjälp av integriner. Den beskriver integriner som binder till två av broskets proteiner CHAD och kollagen typ II. Avhandlingen beskriver också upptäckten av en tidigare okänd kollagenbindande integrin subenhet alpha10, som i huvudsak finns på broskceller. (Less)
