Cellular and Molecular Pathways Governing Hematopoietic Stem Cell Fate

Abstract
Hematopoietic Stem Cells (HSCs) have been defined to have abilities of both self-renewal and multi-lineage differen-tiation. The balance between these abilities is rigorously controlled under steady-state status (homeostasis) and when the normal processes of self-renewal and differentiation become deregulated, a disorder of blood system (such as leukemias) can occur. HSCs are enriched in lineage negative (Lin-), SCA-1 positive and c-KIT positive (LSK) cells in mouse bone marrow (BM), as well as in the Lin(-)CD34(+) cells in human BM or cord blood. HSCs are enriched in lineage negative (Lin-), SCA-1 positive and c-KIT positive (LSK) cells in mouse bone marrow (BM), as well as in the Lin(-)CD34(+) cells in human BM or cord blood. However, the LSK compartment is heterogeneous, containing long-term (LT) and short-term (ST) self-renewing HSCs as well as non-self-renewing multipotent progen-itors (MPPs) and demanding further subfractionation of this population to allow for detailed functional investiga-tions. Even though these different subpopulations are phenotypically and functionally characterized, it is still poorly understood how LT-HSCs make a cell fate decision to differentiate into ST-HSCs and further to MPPs, as well as how the cellular and molecular pathways regulate HSC fate decisions. In this thesis, we identified two distinct subpopulations within BM and fetal liver LSK compartment: ST-HSC population identified as LSKCD34(+/hi)FLT3(-) cells and lymphoid primed multipotent progenitors (termed LMPPs) represented by LSKCD34(+)FLT3(hi) cells. LSKCD34(+/hi)FLT3(-) cells are capable of rapid but short-term reconstitution, high colony-forming units-spleen (CFU-S) and radio-protective activities while LSKCD34(+)FLT3(hi) cells can give rise to lymphoid and myeloid cells but have no self-renewal activity and little or no megakaryocytic-erythrocytic (MkE) potentials. This was further supported by gene expression analysis revealing a modulation of genetic programs in the transition from the ST-HSC to the LMPP, with down-regulation of MkE and up-regulation of lymphoid gene programs. The work has also investigated the regulatory roles of extrinsic modulators on HSC function. We found that HSC self-renewal is negatively regulated by activation of the IFN-gamma and Fas pathways, through the promotion of HSC differentiation. (Less)

Det finns olika slags mogna celler: röda blodkroppar, vita blodkroppar samt blodplättar. Dessa celler är livsnödvändiga. Röda blodkroppar transporterar syre och koldioxid; vita blodkroppar skyddar mot infektioner orsakade av mikro-organismer; blodplättar stoppar en blödning. De flesta av dessa mogna blodceller har en kort livslängd, från några timmar upp till hundra år, vilket gör att de måste nyproduceras kontinuerligt. I normalt tillstånd måste det produceras en miljard blodceller varje dag hos en vuxen individ. Då kroppen utsätts för stress, som akut blodförlust eller infektion, kan blodsystemet snabbt öka produk-tionen av olika blodceller för att bibehålla en jämn nivå av celler i blodet.



Som nämnts ovan upprätthåller blodstamcellerna en kontinuerlig blodbildning. Dessa celler är dock väldigt få i antal, de utgör enbart 0.005-0.01% av hela benmärgen i en mus. Blodsystemet är väldigt dynamiskt, vilket återspeglas i att en enda stamcell kan bilda flera olika mogna blodceller. De omdiskuterade frågorna idag är på vilket sätt utmognaden av blodstamceller sker, d.v.s. vilka faktorer är det som styr om stamcellen kommer att självförnya sig eller producera sin avkomma? Denna avhandling fokuserar på identifiering, karakterisering och reglering av blodstamceller i både mus och människa.



Vårt mål var att identifiera olika subpopulationer inom blodstamcellerna. Våra studier visar att det finns två olika slags kort-tidsstamceller: en kort-tidsstamcell definierad som LSKCD34+FLT3- som kan bilda alla blodkroppar och en annan med övervägande förmåga att bilda vita blodkroppar (kallad LMPP) definierad som LSKCD34+FLT3hi. LSKCD34+FLT3- stamcellerna har förmågan att efter transplantation snabbt repopulera och effektivt rädda letalt bestrålade möss. Dessa egenskaper kan vara väldigt användbara i utvecklingen av protokoll som kan användas i kliniken. LSKCD34+FLT3hi celler har förmågan att bilda granulocyter, makrofager och lymfoida celler, men kan inte längre bilda röda blodkroppar och blodplättar. I tillägg har våra studier undersökt genregleringen i dessa stamceller för att bättra förstå hur stamcellens val att utvecklas till olika blodceller regleras. Vi har också karakteriserat hur tillväxtfaktoerna Fas och interferon kan negativt reglera självförnyelsen av stamceller. Dessa studier hjälper oss att bättre förstå stamcellsbiologi och hur vi därigenom kan använda de erhållna kunskaperna i kliniken för att utveckla metoder för benmärgstransplantation. (Less)
