Structure and distribution of alpha-1-microglobulin proteins

Abstract
This thesis describes studies of the structure and tissue distribution of alpha-1-microglobulin (a1-m), a member of the lipocalin protein superfamily with immunosuppressive properies. It was shown that a1-m appears in human plasma in five major forms. These were identified as monomeric a1-m and covalent complexes between a1-m and IgA, prothrombin, albumin and a1-m. In addition to blood, both monomeric-, and high molecular weight forms of a1-m were present in tissues. The liver, kidney and heart contained a series of a1-m isoforms with apparent molecular masses between 40 kDa and 50 kDa, which were not present in plasma. In addition to blood, both monomeric-, and high molecular weight forms of a1-m were present in tissues. The liver, kidney and heart contained a series of a1-m isoforms with apparent molecular masses between 40 kDa and 50 kDa, which were not present in plasma. Immunohistochemical analysis of human tissue demonstrated intracellular labeling of a1-m in hepatocytes and in the proximal epithelial cells of the kidney. In addition, a1-m immunoreactivity was detected in the interstitial connective tissue of heart, lung and in the adventitia of blood vessels as well as on cell surfaces of cardiocytes. a1-m mRNA was detected only in the liver and pancreas. This suggests that the protein found in other tissues is not synthesized locally, but transported to the tissues via the bloodstream from the production sites in liver and pancreas. a1-m was found in placenta particularly at sites of syncytiotrophoblast injury. Thus, ?tissue a1-m? may act as a local immunosuppressive barrier in the placenta, protecting the chorionic villi from the immune system of the mother. a1-m is yellow-brown. Colored tryptic peptides of a1-m were purified, and it was shown that the side chains of Lys92, Lys118 and Lys130 in these peptides carried size heterogeneous covalently attached groups with molecular masses beetween 106 and 282 Da. The fluorescence properties and molecular modeling suggested that the chromophores were most likely buried and closely spaced in the interior of the protein. Only the free a1m carried colored groups, whereas the a1m linked to IgA was uncolored. (Less)

I denna avhandling har vi undersökt ett brunt protein som heter a1-mikroglobulin (a1-m). a1-m finns dels som fritt protein i blod, men bildar också komplex med andra proteiner. I arbete I har vi tagit reda på vilka andra proteiner som a1-m är bundna till i blod. Resultaten visade att a1-m är bundet till albumin, som är det vanligaste proteinet i blod, samt till protrombin, ett protein som är inblandat i blodkoagulationen (blodlevringen). Det är tidigare visat att a1-m också är bundet till immunoglobulin A (IgA), som är inblandat i kroppens immunförsvar. Dessa komplex har förmodligen en funktion i kroppen, men den är ännu så länge okänd.



a1-m har sedan länge varit känt som ett blodprotein, men inga ordentliga undersökningar har gjorts om den finns i vävnaden utanför blodbanan. Detta undersöks i arbete II. Våra resultat tydde på att a1-m finns nästan överallt ute i vävnaderna, och förmodligen har en viktig funktion där. Vi visade också att a1-m troligtvis inte tillverkas ute i vävnaderna, utan av levern. Det är därför troligt att a1-m transporteras ut till vävnaderna via blodet efter att ha tillverkats i levern.



När immunförsvaret sätts igång, tex. som en respons på en bakterieinfektion, kommer potentiellt farliga processer igång. Bland annat släpps det ut ämnen som bryter ner, inte bara bakterien, utan också kroppens egna proteiner. Det har tidigare visats att a1-m har immunsuppressiva effekter, dvs. det undertrycker kroppens immunförsvar. I arbete III sätter vi upp en hypotes att anledningen till att a1-m hittas i vävnaderna är att det ska skydda dessa vävnader från kroppens eget immunförsvar. För att verifiera denna hypotes undersökte vi om a1-m kunde hittas i placenta. Placentan, eller ?moderkakan? har många mekanismer för att skydda fostret från moderns eget immunförsvar. Om dessa mekanismer ej hade funnits hade barnet känts igen som främmande, icke kroppseget, material som måste stötas bort. Våra resultat tydde på att a1-m finns i ganska stora mängder i placentan, lokaliserat bl.a. till syncytiotrofoblasterna. Syncytiotrofoblasterna utgör det cell-lager som ligger närmast moderns blod och bildar därigenom gränsytan mellan fostret och modern. För att utöva maximalt skydd mot moderns immunförsvar finns det flera mekanismer som undertrycker immunförsvaret just i syncytiotrofoblasterna. Om det blir en skada på syncytiotrofoblasterna exponeras andra delar av placentan som inte har de skyddande mekanismerna. Dessa kan då attackeras av moderns immunförsvar. Intressant nog, tydde våra resultat på att a1-m finns i extra stor mängd just där det har blivit en skada på synsytiotrofoblasterna. Man kan alltså tänka sig att a1-m binds in till de skadade delarna och där undertrycker modens immunförsvar, så att fostervävnaden inte tar skada. Vi visade också att det finns flera former av a1-m i placenta som ser annorlunda ut än i blod.



a1-m tillhör en proteinsuperfamilj som kallas lipokaliner. Alla lipokaliner har liknande struktur: Aminosyrakedjan är hopvikt på ett sätt som får dessa proteiner att likna ett kafferfilter, med en centralt belägen ficka. Många lipokaliner transporterar olika ämnen i ?kafferfilterfickan?. Ett exempel på detta är retinol-bindande protein som bär på en viss form av vitamin A bundet i fickan. Som tidigare nämnt är a1-m brunfärgat. I arbete IV visade vi att det bruna ämnet förmodligen är bundet just i fickan på a1-m. Vi visade också på vilka aminosyror som den bruna föreningen sitter, samt mätte exakt dess vikt. Bara fritt a1-m verkade bära på den bruna gruppen. a1-m som är bundet till IgA (se arbete I) var ofärgat. Anledningen till detta är inte känd, men det är möjligt att av och påsättning av den bruna föreningen regleras med hjälp av komplex bindning med andra proteiner, tex. IgA. (Less)
