Abstract
T cells constitute a heterogenous population of lymphocytes. Their unifying property is that they express surface bound antigen-receptors, called T cell receptors (TCRs). The TCR is a heterodimer composed of either TCRalpha and beta chain proteins (alphabeta TCR) or gamma and delta chain proteins (gammadelta TCR). Depending on which TCR isotype that is expressed, the T cells are divided into alphabeta T cells or gammadelta T cells. T cells constitute a heterogenous population of lymphocytes. Their unifying property is that they express surface bound antigen-receptors, called T cell receptors (TCRs). The TCR is a heterodimer composed of either TCRalpha and beta chain proteins (alphabeta TCR) or gamma and delta chain proteins (gammadelta TCR). Depending on which TCR isotype that is expressed, the T cells are divided into alphabeta T cells or gammadelta T cells. In contrast to other hematopoietic cells, T cells develop in an organ called the thymus. To become a mature T cell the thymocytes proceed through a differentiation program that is partially induced by the surrounding thymic epithelial cells. T cell development can be divided into an early and a late stage of differentiation. The purpose of the early stage is to provide the developing thymocytes with functional TCRs. This is mediated by rearrangements at the DNA level of the TCRbeta, gamma and delta genes. If the TCRbeta gene is functionally rearranged, it will pair with a protein called pTalpha to form the preTCR complex that assures rearrangements of the TCRalpha genes and differentiation into the alphabeta T cell lineage. On the other hand, if the TCRgamma and delta chain genes are rearranged functionally the thymocyte will deviate from the mainstream alphabeta T cell pathway to become a mature gammadelta T cell. Whereas conventional alphabeta T cell development is quite well understood today, that of unconventional alphabeta T cells and gammadelta T cells is not. This thesis concerns the early stages of T cell development. With the articles herein, we provide information about the role of the TCR and preTCR in alphabeta versus gammadelta T cell development. We have furthermore examined the expression and transcriptional regulation of the pTalpha gene. Together, these findings lead to a better understanding of the early events in T cell development. De sistnämnda kan bidra till att en inflammation och feber uppstår, men de kan även presentera bitar av patogenerna (antigen) för det specifika immunförsvaret på antigen presenterande celler (APCs). Cellerna i det specifika immunförsvaret kallas lymfocyter och består av B och T celler. Efter att ha aktiverats av APCs producerar B cellerna antikroppar som är specifika mot det aktuella antigenet. T cellerna aktiveras också, men det finns flera typer av T celler med olika funktioner. Vissa har som funktion att döda infekterade celler direkt med specificitet (cytotoxiska T celler; Tc) andra att styra försvaret mot ett Tc svar eller B cells svar (T hjälparceller; TH1 eller TH2). Lymphocyterna kan även bilda minnes celler som kan aktiveras snabbt och effektivt nästa gång samma antigen dyker upp (principen bakom vaccin).



Den här avhandlingen behandlar tidig T cells utveckling. Förutom de ovan nämnda T cellerna finns det även små undergrupper som man skiljer på med hjälp av deras yt-proteiner, deras funktion och var de befinner sig anatomiskt. På cellytan har T celler ett proteinkomplex som kallas T cells receptorn (TCR), vilken har som funktion att känna igen olika antigen. Varje T cell har en specifik T cells receptor som känner igen enbart ett antigen. T cells receptorn har en enorm diversitet och därför kan vår kropp bekämpa nya patogener så effektivt. T cells receptorn finns i två olika former bestående av alphabeta eller gammadelta TCR proteiner och beroende på vilken T cells receptor som används delar man in T cellerna i alphabeta T celler och gammadelta T celler. Till alphabeta T cellerna hör de ovan nämnda TH och Tc cellerna vilka återfinns framförallt i de perifera organen (mjälte, lever, lymfnoder etc; 95% av perifera T cellerna). gammadelta T cellerna finns framförallt i de epiteliala lagren (tarm, hud, slemhinnor etc; 50-99%) men även perifert (1-5%). Dessa T celler skiljer sig mycket från de konventionella alphabeta T cellerna. Dessutom finns en liten population alphabeta T celler som även uttrycker NK cells markörer och dessa kallas därför NKT celler.



T cellerna bildas i ett organ som heter tymus. I tymus genomgår de en utvecklingsprocess som omfattar en negativ selektion (eliminering) av de T celler som uttrycker en själv-specifik T cells receptor. Om dessa inte elimineras kan man få auto-immuna sjukdomar. De omogna T cellerna kallas för tymocyter och härrör från början från hematopoietiska stamceller i benmärgen. Det finns rätt så god kunskap om hur alphabeta T cellerna bildas från en tymocyt, men vi vet mindre om hur gammadelta T celler och NKT celler bildas och framförallt om hur en tymocyt bildas från en stamcell. I denna avhandling redogör jag för fältet "tidig T cells utveckling", vilket innefattar de första stegen i en tymocyts utveckling såsom när den bestämmer sig för vilken sorts T cell den ska bli. Det är allmänt känt att en och samma tymocyt har potential att bli alla sorters T celler, men man vet inte riktigt hur. Vi har studerat hur okonventionella alphabeta T celler (DN alphabeta T celler) och gammadelta T celler bildas. Dessutom har vi undersökt uttrycket och regleringen av ett oumbärligt protein för konventionell alphabeta T cells utveckling, pTalpha. Artiklarna som ligger till grund för denna avhandling har bidragit till ökad förståelse av T cell receptorns roll i T cellens utveckling och gett oss viktig molekylärbiologisk kunskap om hur pTalpha genen regleras.
