We have performed a series of experiments in order to elucidate the properties of two breast cancer related microRNAs, here we denote A and B.
In one experiment, we tested varying concentrations of biologically relevant cation upon the melting curve of A. We saw that the monovalent conditions favoured a melting curve that had two inflexion points whereas the divalent conditions favoured only one inflexion point. This informs us on how the ionic conditions of the microRNA environment affect its thermostability as well as hint at RNAs tendency to form secondary structures.

We saw that the monovalent conditions favoured a melting curve that had two inflexion points whereas the divalent conditions favoured only one inflexion point. This informs us on how the ionic conditions of the microRNA environment affect its thermostability as well as hint at RNAs tendency to form secondary structures.
Another path saw us study B wild type (WT), as well as B mutant. In vitro studies allowed us to find the various energies associated with the microRNAs, with the mutant having a ΔG° of 52 kJ mol 1 and the WT of 71 kJ mol 1. The smaller ΔG° for the mutant indicates it is less stable then the wildtype.
The in vivo luciferase studies looked at B WT and mutant and their propensity for down regulation of Firefly luciferase activity. The Firefly reporter gene was present under the influence of the 3’-UTR BRCA1, in a pEZX-MT01 transfected in to MCF-7 cancer cells. The results showed a decreased activity of the Firefly luciferase in regard to the WT but the mutant had no effect. This coincides with current knowledge on B targets and method of downregulation. Kontinuerlig translation av protein som inte behövs av cellen är ett slöseri med resurser och kommer att leda till celldöd. Olika kontrollmekanismer i cellen förhindrar detta. En sådan kontrollmekanism är nyttjandet av små RNA-molekyler, kallade mikro-RNA (även miRNA), för att reglera hastigheten på proteinsyntesen.
Detta sker genom att mänskligt miRNA binder till en icke-translaterad region av budbärar-RNA:t. Konsekvensen av detta är en minskning av translationshastigheten vilket resulterar i färre proteiner syntetiserade per budbärar-RNA. Det finns också en möjlighet att budbärar-RNA:t förstörs, vilket resulterar i att inga nya proteiner längre kan syntetiseras från det.
Forskarna har kunnat kopiera miRNA-mekanismen via syntetiska alternativ i en process kallad RNA-interferens (RNAi), med hjälp av små interferande RNA (siRNA). Detta är ett växande forskningsfält och har mycket lovande förutsättningar för behandling av genetiska sjukdomar och cancer. Effekten av befintliga cancerbehandlingar på siRNA och dess effektivitet är okänd. Vissa platinumbaserade föreningar, som exempelvis läkemedlet cisplatin, vet man att de interagerar med DNA och de olika RNA-typerna. Resultaten beskrivna i denna rapport visar på förändringar i termodynamisk stabilitet hos utvalda miRNA i förhållande till cisplatin och andra biologiskt relevanta ämnen. Vår slutsats är att förändringar i RNA eller dess miljö förändrar beteendet hos miRNA:t jämfört med förväntade trender.
