However, these receptors are prone to be affected by the digestion enzymes used to obtain single cell suspension from primary tissue. In this study, we investigated the DC yield as well as the expression of several CLRs on DCs after treatment with two commonly used digestion protocols; a protocol commonly used within the field of DC research (Standard Enzyme protocol) as well as a commercially available kit, compared to a control where no enzymes were applied. Single cell suspension of tonsillar tissue, as well as peripheral blood mononuclear cells (PBMCs), was stained with antibody (Ab) panels and cell surface expression levels of various CLRs was analyzed using flow cytometry. The results demonstrated reduced expression of CLR surface receptors CLEC9a, DEC205, CLECSF14 and Dectin 1 using the Standard Enzyme protocol but not when using the commercial kit. The yield was not affected for CD141+ and CD1c+ DC between the different protocols but a decrease was seen for CD123+ DCs when using the Standard Enzyme protocol in comparison to the Control protocol. This study stresses the importance of selecting the appropriate protocol for the downstream analysis and suggests that previous studies on these CLR should be interpreted in relation to the enzymatic degradation applied. Denna strategi fungerar genom att ett virus kopplas till den skräddarsydda antikroppen som sedan hittar de dendritiska cellerna. Istället för att immunförsvaret ska hitta viruset så är det lite som hemkörning - viruset kommer till dem.
För att skapa en antikropp som är specialdesignad för att hitta dendritiska celler måste vi veta mer om dendritiska celler. Dendritiska celler kan i sin tur delas upp i flera grupper så kallade subtyper där varje subtyp har specialiserat sig på en unik uppgift, en subtype har specialiserat sig på att ta hand om virus medan en annan är bättre på att ta hand om tumörceller.
I mitt examensarbete har jag tittat på vilka proteiner som finns hos olika dendritiska subtyper så att dessa kan urskiljas från andra dendritiska celler. En typ av proteiner som ofta hittas på ytan hos dendritiska celler är pattern recognition receptors som förkortas PRR. Dessa proteiner är de som bland annat kan känna igen om organismer/celler ska tolereras eller bekämpas. Det finns PRRs som bara känner igen virus och andra som bara känner igen tumörer.
Proteiner är väldigt känsliga och bryts lätt ner. Jag har jämfört olika protokoll som använts inom forskningsfältet för att få ut dendritiska celler och subtyper för att studera dem och det visade sig att de kan inverka på vilka proteiner som kan mätas på ytan. Det jag kommit fram till är att användningen av enzymer vid isoleringen av dendritiska celler kan bryta ner proteinerna på ytan av dendritiska celler och dess subtyper. Därför är det väldigt viktigt att välja rätt enzymer vid isoleringen.
Genom att ha en bättre förståelse vilka proteiner som finns på ytan hos dendritiska celler kan en specialdesignad antikropp skapas. Den kan sen användas som en målsökare för att hitta de dendritiska cellerna och dess subtyper var de än är i kroppen. Nästa steg är att koppla ett virus eller en tumörbit till antikroppen. Så att viruset eller tumörbiten kommer direkt till de önskade dendritiska subtyperna. De dendritiska subtyperna lär sig i processen att känna igen viruset som något dåligt och säger till resten av immunförsvaret att skapa immunitet mot viruset eller tumören. (Less)
