- Mantle cell lymphoma (MCL) is an aggressive B-cell lymphoma with poor response to generalized treatment regimens. - Mantle cell lymphoma (MCL) is an aggressive B-cell lymphoma with poor response to generalized treatment regimens. Tools for stratifying patients based on molecular profiles of the tumor, such as the expression level of a biomarker, are needed to enable more targeted therapies. A potential biomarker for MCL patient stratification is the surface protein CD25. This study aimed to assess the frequency and biological impact of B- and T-cell CD25 expression in MCL, and to investigate its prognostic impact in relation to established risk factors and survival. The morphology-, proteomic- and transcriptomic profiles of MCL tissue samples were investigated using the digital spatial profiler GeoMx from Nanostring. The study showed that the majority (90%) of the total CD25+ population (sum of CD25+ B-cells and T-cells) in our patient cohort constitutes B-cells. In addition, our results indicate a constitutive malignant CD25 expression. Differentially expressed genes and proteins were identified between patients with high and low CD25 expression when comparing both B- and T-cell mRNA and protein profiles. No significant correlation between CD25 expression and survival was observed. Furthermore, our results indicate that a high expression of CD25 is significantly associated with a low Ki-67 index and non-mutated TP53 gene, thus suggesting it to be associated with a favorable subtype of the disease. However, more studies are necessary to be able to draw conclusions about the biological relevance of malignant CD25 expression in MCL, and its potential to be used for patient stratification. (Less)

Ett sätt att utvärdera potentialen hos en biomarkör är att undersöka dess eventuella koppling till överlevnad och till faktorer som kan påvisa ett visst sjukdomsförlopp, så kallade prognostiska faktorer. Jakten på biomarkörer som kan indikera ett mer precist tumörtillstånd pågår ständigt, och är central i utvecklingen av riktade cancerterapier.
Syftet med detta projekt var att studera en potentiell biomarkör i mantelcellslymfom och hur uttrycket av denna hos cancerceller påverkar patientens prognos och överlevnad. Överuttryck av biomarkören har tidigare hittats i andra relaterade cancertyper, vilket gör den intressant att undersöka i mantelcellslymfom. Genom att analysera vävnadsprover samt bilder av dessa vävnadsprover från patienter diagnostiserade med mantelcellslymfom har denna studie visat att det finns tumörceller som uttrycker biomarkören och generellt sett så har patienter antingen väldigt många eller ytterst få sådana cancerceller. Vi kunde inte påvisa något samband mellan uttrycket av biomarkören och överlevnad. Däremot kunde vi se en koppling till två olika prognostiska faktorer som indirekt skulle kunna visa på att ett högt uttryck av markören är kopplat till en subgrupp av tumörer med mer fördelaktig prognos.
Biomarkören som vi undersökt har visat sig vara kopplad till överlevnad i flera andra cancertyper och därför finns det anledning att tro att den har potential även i mantelcellslymfom. Vår studie kan ses som en början på forskningen om denna biomarkör men fler studier krävs för att kunna dra slutsatser om dess biologiska betydelse i mantelcellslymfom och framförallt om dess använding som en markör för att indikera subtyper av tumörer.
