Abstract
This thesis describes early inflammatory responses involving interactions between leukocytes and the endothelial lining of the blood vessel wall. An adhesion molecule, E-selectin, which is upregulated on interleukin-1 activated endothelium, was found to mediate rolling between leukocytes and endothelial cells in vivo. Immunohistochemistry revealed that E-selectin was preferentially expressed in venules and not in arterioles. This thesis describes early inflammatory responses involving interactions between leukocytes and the endothelial lining of the blood vessel wall. An adhesion molecule, E-selectin, which is upregulated on interleukin-1 activated endothelium, was found to mediate rolling between leukocytes and endothelial cells in vivo. Immunohistochemistry revealed that E-selectin was preferentially expressed in venules and not in arterioles. In most inflammatory conditions, leukocyte rolling and subsequent extravasation to surrounding tissues take place in the postcapillary venules which is also the site for inflammation mediated vascular permeability. We found that a gradient of the inflammatory mediator, LTB4, was essential for the induction of neutrophil-mediated increased permeability and migration. Furthermore, permeability was found to be increased by the cationic and neutrophilic granular protein heparin binding protein (HBP, CAP37, azurocidin). The effects of HBP on endothelial barrier function could be partially neutralized by anti-HBP antibodies. In addition, HBP was shown to increase intracellular Ca2+-levels and rearrange actin-filaments in endothelial cells which showed that HBP was capable of inducing the cellular changes that are required for cell contraction and intercellular gap formation. Affinity chromatography of HBP binding molecules showed that HBP bound to all known endothelial cell surface proteoglycans. Flow cytometry and microscopical investigations demonstrated that HBP was rapidly internalized into endothelial cells. Subcellular fractionation, confocal microscopy and electron microscopy studies led us to the conclusion that HBP was localized to the mitochondrial compartment. Addition of HBP to endothelial cells prior to the induction of apoptosis, by growth factor withdrawal, showed that HBP protected the endothelial cells from apoptosis. HBP was also shown to interact with a mitochondrial resident protein, p33, and to decrease incorporation of 32PO4 into endothelial proteins. (Less)

I min avhandling har jag studerat hur neutrofilerna hittar till rätt plats i kroppen, vad de gör för att lämna blodet och hur vi tror att de gör för att angripa de infekterande mikroorganismerna så effektivt som möjligt utan att skada kroppens friska celler.



När de inflammatoriska mediatorerna når endotelcellerna, som fullständigt täcker insidan av våra blodkärl, ändrar de sin cellyta och blir "klibbiga". När neutrofilerna förs med blodet saktar de ned farten och börjar rulla på insidan av blodkärlet, där de normalt sett bara skulle åka förbi. Ju starkare inflammationen är desto långsammare rullar de för att till sist fastna och stanna. Vi har funnit att E-selektin som finns på endotelcellerna är viktigt för att neutrofilerna skall kunna rulla. E-selektin är ett cellyteprotein, dvs ett äggviteämne som sitter förankrat i cellen. När vi tillsatte antikroppar mot E-selektin rullade neutrofilerna sämre och fick därmed svårare att stanna.



När neutrofilerna har fastnat utsöndrar de signaler som gör att endotelcellerna särar lite på sig. Därmed sipprar vätska ut från blodkärlen. Processen kallas läckage men är en noggrant reglerad process. De proteiner som läcker ut från blodet underlättar oskadliggörandet av mikroorganismer i den omkringliggande vävnaden. Neutrofiler kan utsöndra ett stort antal molekyler för att bekämpa infektioner, men vilken molekyl som är ansvarig för att starta läckage har inte varit känd. Vi har studerat Heparin Binding Protein, HBP, i flera olika försöksuppställningar. HBP kan inducera läckage både vid in vivo och in vitro försök, dvs både i djur och i cellodling. Vi studerade endotelcellerna efter tillsats av HBP och såg att deras cellskelett förändrades vilket kan vara en del av förklaringen till hur de särar på sig.



Efter att neutrofilerna har fastnat går de ut i vävnaden, där de fortsätter att utsöndra molekyler som ger signaler till andra celler eller är giftiga för mikroorganismer. Tidigare har man visat att HBP binder till en komponent av mikroorganismer och ser till att de oskadliggörs. Vi ville studera om HBP hade ännu fler effekter. Vi fann att HBP binder till proteoglykaner på endotelceller. Proteoglykaner består av många socker molekyler i kedjor som sitter fast på en gemensam proteinkedja. Vi studerade hur endotelceller tog upp HBP från sin omgivning och noterade att proteoglykaner delvis var involverade i denna process. HBP fördes sedan vidare till mitokondrierna inuti cellen. Mitokondrier är cellens kraftstation. Den använder delar av socker för att göra energipaket som cellen kan använda. Celler som inte får näring eller skadas på annat sätt begår ett rituellt självmord och man vet nu att mitokondrierna har en aktiv roll vid denna självdestruerande process. Här har vi visat att HBP skyddar endotelceller mot den naturliga celldöden vilket vi tror har betydelse för läkningsprocessen vid inflammationer. (Less)
