Modeling and Calibration of Preparative Chromatography

- In my thesis, I have modeled adsorption chromatography and simulated, calibrated, and analyzed models of adsorption chromatography.
While my main focus has been modeling and calibration of the models by estimating the parameters, the models used have been partly new.
The simulations are differentiated from the modeling in that the modeling describes the partial differential algebraic equation, while the simulation describes the discretization and solution of the model.
When a model is calibrated, the model parameters are estimated to replicate experimental data. I have developed a method to find how model parameters may depend on experimental errors.
The process I have worked most with is ion-exchange chromatography, operated in bind and elute mode, and the model I have used most to describe the adsorption process is the steric mass action model.

produkt som faktiskt når marknaden måste därför täcka kostnaden, inte bara

läkemedelspatent varar i ett antalår. För att täcka utvecklingskostnaderna är det
därför viktigt att produkten når marknaden så snabbt som möjligt.
Före en storskalig process framställsningsprocess av läkemedlet är designad,
finns det en begränsad mängd av det. Genom att anpassa en matematisk modell
till ett fåtal experiment kan man förutsäga vad som troligen skulle hända om man
gjorde andra experiment. Fördelen är att efter att modellen har blivit anpassad
går det inte åt mer läkemedel och en simulering får ett svar mycket snabbare
än från experiment. Detta gör att vi kan testa en mycket stor mängd möjliga
processalternativ för att hitta något som verkar lovande. De kombinationer som
verkar lovande kan sedan valideras med experiment.
Vi kan även utvärdera vad som händer med processen om den utsätts för
störningar. I mitt fall är det mest effekter av att processen varierar som unders
ökts. Det kan till exempel innebära att temperaturen inte behöver vara exakt
samma varje gång, utan kan variera lite som den ofta gör i en fabrikslokal. Jag tittar
också på vad som händer om den storskaliga processen inte är lika kontrollerad
som ett experiment i ett laboratorium. Om produkten inte är tillräckligt ren kan
läkemedelsföreget inte sälja det de har producerat. Det innebär en förlust i både
tid och resurser. Om det finns en risk för att mängden föroreningar blir för hög kan
lösningar på hur denna risk minimeras föreslås. Med modellen kan man utvärdera
vilken effekt det får att till exempel temperaturen är lite högre och att saltkoncentrationen
är lite lägre än vad som vore optimalt. Man kan förutsäga hur mycket
mer föroreningar produkten kommer innehålla och hur mycket av produkten som
kommer gå förlorad. (Less)
