- Generation of long-lived plasma cells and memory B cells of high affinity takes place in germinal centers and depends on help from a specialized CD4+ T cell subset termed T follicular helper (Tfh) cells. These processes are crucial for long-term protection upon natural infection or in settings of vaccination, however they are also responsible for development of antibody-mediated autoimmune disease.
Targeting of Tfh cells may therefore be of importance both in the development of new vaccines and in treatment of autoimmune conditions associated with pathologic autoantibody production. These processes are crucial for long-term protection upon natural infection or in settings of vaccination, however they are also responsible for development of antibody-mediated autoimmune disease.
Targeting of Tfh cells may therefore be of importance both in the development of new vaccines and in treatment of autoimmune conditions associated with pathologic autoantibody production. The aim of this thesis was to examine the molecular and cellular interactions in the generation and function of Tfh cells.
First we wanted to determine if CXCR5+ Tfh cells are developmentally distinct from peripheral gut homing Th cells, and if they differ in terms of lymph node egress.
We could observe that CXCR5+ Tfh cells and α4β7+ gut-homing Th cells are generated as two separate subsets and that majority of CXCR5+ Tfh cells were retained within the lymph node whereas the majority of α4β7+ Th cells exited.
Second we looked at the role of type I interferon and dendritic cells in the generation and function of Tfh cells. We observed that type I interferon signaling in DCs is important for development of Tfh cells in response to antigen and TLR3 or TLR4 agonists and suggested that it functioned by inducing IL-6 production. We have also demonstrated that conventional CD11c DCs at least partially through their robust co-stimulation via CD28 provide unique signals essential for development of functional Tfh cells.
A deeper knowledge of these processes would undoubtedly be valuable for the development of new vaccines as well as for advances in treatment of autoimmune diseases. (Less) - Abstract (Swedish)

Immunförsvaret delas in i det medfödda och det adaptiva. Det medfödda
immunförsvaret utgör den första försvarslinjen vid en infektion medan det
adaptiva behöver några dagar för att aktiveras. Dendritceller och makrofager är två
olika typer av vita blodkroppar som tillhör det medfödda immunförsvaret. Dessa
känner igen strukturer som är gemensamma för många olika mikroorganismer. När
dessa kommer i kontakt med t. ex. en bakterie som invaderat ett sår, kommer de att
producera olika ämnen som attraherar andra immunceller till infektionsplatsen.
Om infektionen kvarstår, aktiveras det adaptiva immunförsvaret, en process som
sker i lymfkörtlarna. T celler och B celler är vita blodkroppar och är de viktigaste
komponenterna i det adaptiva immunförsvaret. Till skillnad från det medfödda
immunförsvaret, är det adaptiva mycket mer specifikt, dvs. varje enskild T cell och
B cell är unik och kan känna igen små förändringar hos ett antigen. T och B celler
som inte tidigare träffat på ett antigen, vandrar ständigt mellan blodcirkulationen
och lymfkörtlarna. Dendritceller som vid infektionsstället tar upp antigen kommer
att vandra via lymfkärl till lymfkörtlarna där de presenterar det för T cellerna. Om

dessa känner igen antigenet aktiveras de och utvecklas till effektor T celler. Till
skillnad från T celler känner B cellerna igen fria antigen som transporterats från
infektionsstället via lymfan in till lymfkörtlarna. T och B celler interagerar sedan
med varandra för ytterligare aktivering varpå de börjar dela sig för att effektivt
kunna bekämpa infektionen.
CD4+ T celler som jag har studerat är en undergrupp av T celler och kan delas in i
en central och perifer grupp beroende på vilken funktion de har för vårt
immunförsvar. Perifera CD4+ T celler vandrar till infektionsstället efter att de
genomgått aktivering i lymfknutorna där de hjälper andra celler med att bekämpa
infektionen medan de centrala förblir kvar i lymfknutorna och stimulerar B celler
att producera antikroppar. Om perifera och centrala T celler utgör helt skilda
populationer, eller om de delvis är överlappande, har inte fastställts.
Tidigare data visar att perifera CD4+ T celler som migrerar in i tarmen uttrycker
adhesionsmolekylen α4β7 som styr T cellens vandring in i tarmslemhinnan. Vi har
tittat på en grupp av CD4 T celler som kallas för follikulära T hjälparceller (Tfh).
Dessa uttrycker receptorn CXCR5 vars ligand uttrycks i B cellsrika regioner i
lymfknutorna. På detta sätt hittar de till B cellerna varefter de stimulerar dessa att
producera högaffina antikroppar mot den specifika infektionen. Högaffina
antikroppar är ofta mycket effektiva på att neutralisera skadliga virus, bakterier
och toxiner. Tfh cellernas centrala roll i regleringen av denna
antikroppsproduktion understryker därför hur viktiga dessa celler är för vårt
immunförsvar. Högaffina antikroppar har även påvisats i en rad autoimmuna
sjukdomar och det tycks som om att Tfh celler kan vara involverade även i
utvecklingen av autoimmunitet.

Vi har tittat på CD4+ T celler i lymfknutorna i tarmen och visat att där bildas två
separata icke överlappande populationer av CXCR5+α4β7
- och α4β7
+CXCR5- T
celler. Dessutom har vi visat att de CXCR5+α4β7
- Tfh cellerna selektivt förblir
kvar i lymfknutan. Vi har även visat att dendritceller samt typ I interferoner,
proteiner som produceras vid en infektion, är nödvändiga för utvecklingen av Tfh
celler. Att studera mekanismer bakom utvecklingen av dessa celler är värdefullt
för att i framtiden kunna utveckla nya vacciner, samt finna bättre behandling eller
helst bota autoimmuna sjukdomar. (Less)
