Specific monitoring of chromatographic procedures

Abstract
On-line monitoring of chromatographic procedures involving biomolecules is usually restricted to UV registration and determination of the pH or conductivity of the mobile phase. A more specific monitoring of proteins, for instance direct identification of a special enzyme, would greatly facilitate and speed up purification. We have found that flow calorimetry, performed with an enzyme thermistor instrument as the detector of enzymic activity, has definite advantages. Moreover, the same instrument can be utilized in an automated, rapid on-line thermometric immunoassay for biomolecules other than enzymes.
