Aqueous solid-phase peptide synthesis (ASPPS) approaches to complex peptides

Abstract
The most prevalent method of peptide production, solid-phase peptide synthesis (SPPS), generates large amounts of hazardous solvent waste, typically DMF. This waste, combined with the use of the insidious PFAS chemical TFA for side chain PG removal and final resin cleavage, has prompted research into PFAS-free and aqueous solid-phase peptide synthesis (ASPPS) of linear peptides. With the growth of branched chain GLP-1 medications, ASPPS exploration of complex peptides is essential.

The target peptide was successfully synthesized with a purity of 40%, demonstrating the feasibility of fully aqueous and PFAS-free synthesis of ε-Lys branched peptides in ASPPS. (Less)

Speciellt har de så kallade GLP-1 läkemedlen, som Ozempic, visat sig vara en mycket effektiv behandling för både diabetes och övervikt. Den huvudsakliga metoden att producera peptider, fast-fas-peptid syntes (solid phase peptide synthesis, SPPS), utvecklades under 1960-talet och bygger på stegvis koppling av aminosyror till ett fast material. Speciellt har de så kallade GLP-1 läkemedlen, som Ozempic, visat sig vara en mycket effektiv behandling för både diabetes och övervikt. Den huvudsakliga metoden att producera peptider, fast-fas-peptid syntes (solid phase peptide synthesis, SPPS), utvecklades under 1960-talet och bygger på stegvis koppling av aminosyror till ett fast material. SPPS medför dock stora mängder av miljö- och hälsofarliga lösningsmedel samt PFAS-kemikalien TFA. Det är därför viktigt att utveckla metoder med mindre mängd lösningsmedel och ersätta miljöfarliga lösningsmedel med mer miljövänliga alternativ, till exempel vatten. Fokus inom detta område har varit syntes av linjära peptider men för att skapa GLP-1 mediciner krävs även möjlighet till grenade peptider.
Målet för detta projekt var därför att använda vattenbaserad fast-fas-peptid syntes för att producera en mindre peptid som innehåller sidokedjan för GLP-1 peptiden tirzepatide, samt klyva denna peptid utan PFAS-kemikalier. Först syntetiserades en linjär peptid där aminosyran lysins sidokedja var skyddad med den kemiska gruppen pNZ. Sedan utvärderades betingelser där sidokedjan bäst kunde kopplas på.
Målpeptiden syntetiserades med en renhet på 40% vilket visar att det är möjligt att skapa lysin förgrenade peptider med enbart vatten som lösningsmedel. Detta resultat kan användas som underlag för kommande forskning inom vattenbaserad SPPS av fullständiga GLP-1 peptider. (Less)
