- Amyloid fibrils are aggregates of self-assembled peptides and proteins that forms a highly ordered and densely packed core of β-strands stacking perpendicular to the length of the fibril. Amyloid fibrils of the protein α-synuclein (α-syn) are associated with Parkinson’s disease. This study aims at contributing to the physiochemical basis of α-synuclein aggregation and dissolution, mainly in terms of reaction rate, rate constants and solubility. - Amyloid fibrils are aggregates of self-assembled peptides and proteins that forms a highly ordered and densely packed core of β-strands stacking perpendicular to the length of the fibril. Amyloid fibrils of the protein α-synuclein (α-syn) are associated with Parkinson’s disease. This study aims at contributing to the physiochemical basis of α-synuclein aggregation and dissolution, mainly in terms of reaction rate, rate constants and solubility. We will use 10 mM MES buffer at pH 5.5 and measure ThT and OPA fluorescence intensity to monitor changes in fibril mass over time and free monomer concentration, respectively. We found that amyloid fibrils of α-syn dissolve at 7 ºC and that the dissolution rate increases with dilution and with increasing concentration of NaCl. Furthermore, NaCl and CaCl2 induces dissolution at 37 ºC. However, the dissolution rate seems to decrease with salt concentration and the change in free monomer concentration indicates that the decrease in ThT fluorescence intensity we observe may be a change in morphology instead of loss of total fibril mass. The solubility of α-syn at pH 5.5 is around 3,0 µM. Moreover, ThT have a positive kinetic effect on aggregation, but does not affect solubility. We also found that the dissolution of α-syn amyloid fibrils is reversible. (Less)

α-Synuklein (α-syn) är ett protein vars amyloida fibriller är relaterade till Parkinsons sjukdom. I denna studie har vi försökt hitta förhållanden som leder till upplösning av α-syn fibriller och mätt dess hastighetskonstanter och löslighet. Genom att använda sig av fluoroforerna ThT och OPA kan man få fram värden på upplösningshastighetskonstanter respektive löslighet.
Vi observerade att fibrillerna löstes upp i låg temperatur (7 ºC), d.v.s. fibrillmassan minskade och lösligheten ökade. I närvaro av salt i låg temperatur ökade hastigheten på upplösningen. Ju mer salt som tillsattes, desto snabbare löstes fibrillerna upp. Däremot minskade fibrillmassan lika mycket både med och utan salt, vilket betyder att salt endast påskyndade upplösningen.
Fibrillerna bildades vid 37 ºC och förväntas därför inte lösas upp vid denna temperatur. Vid tillsatts av salt till fibriller i 37 ºC verkade det dock som om fibrillmassan minskade. Det skulle även innebära en ökad löslighet, vilket inte var fallet. Vi misstänker därför att fibrillerna har lösts upp, men istället återbildat fibriller med en annan struktur. Även om dessa nya fibriller bildar samma totala mängd fibrillmassa är det möjligt att vår mätning uppfattar det som en minskning av fibrillmassa. (Less)
