On chip integrated sample preparation for proteomics

- The aim of this thesis project has been to create miniaturized analytical tools that improve sample preparation for the very rapidly growing field of proteomics research. The presented miniaturized analytical tools provide novel and generic analytical solutions, which are highly adaptable for analysis of various low abundant biomolecules. The adaptability and generality of the analytical technology is very important since there are a great diversity both in the analysis methods used and the biomolecules themselves. - The aim of this thesis project has been to create miniaturized analytical tools that improve sample preparation for the very rapidly growing field of proteomics research. The presented miniaturized analytical tools provide novel and generic analytical solutions, which are highly adaptable for analysis of various low abundant biomolecules. The adaptability and generality of the analytical technology is very important since there are a great diversity both in the analysis methods used and the biomolecules themselves. This technology is achieved through the use of silicon micromachning. The thesis presents microstructure developments both in the form of improvements of existing technology and new hardware in the form of microstructures for sample preparation purposes in proteomics. This is achieved by integrated on-chip sample preparation enabling enrichment and sensitive analysis by mass spectrometry.
A sample preparation platform (spot-on-a-chip) consisting of a microdispenser and nanovial arrays enables efficient protein enrichment and sensitive microspot sample preparation for Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) analysis is presented. Technology for improved enzymatic digestion and highly efficient solid-phase microextraction is also demonstrated. (Less) - Abstract (Swedish)

stor. Utveckling av mikroteknologi som tillåter snabba och billiga storskaliga studier
av proteomet och dess koppling till genomet kommer därför att vara essentiell för
den medicinska och farmakologiska forskningens framsteg. Dessa forskningsframsteg
kommer att medföra utveckling av diagnostik och läkemedel för behandling av
många idag svåra eller obotliga sjukdomar (Alzheimers, Cancer, m.fl.). Proteiner
och peptider är involverade i alla funktioner och aktiviteter i levande celler. Ur
farmaceutisk synvinkel är de proteiner/peptider som är inblandade i olika signaloch
regulativa processer i cellerna särskilt intressanta. Analys av proteiner kan ge
både diagnostisk information och information om substans effektivitet. Det finns
därför ett starkt intresse inom både biokemi, cell och molekylär biologi och den
farmaceutiska industrin för utveckling av analyssystem som tillåter snabb och känslig
analys av protein-/peptidprover. Då dessa prover oftast har väldigt liten volym
(1-10 µl) så är mikrostrukturer i kombination med masspektrometri en idealisk
kombination.
I de presenterade publikationerna visas hur proteomikanalys kan förbättras genom
kombination av mikroteknologi och masspektrometri (MALDI-MS). Den
presenterade mikroteknologin tillåter analys med högre känslighet och mindre
provvolymer än vad som är möjligt med konventionella metoder. (Less)
