Abstract
The mechanism of gallbladder contraction induced by infusion of hydrochloric acid (HCl) into the duodenum in dogs with chronic Jones gallbladder cannula, Thomas pancreatic fistula and gastric fistula was investigated. The results were as follows:
1. Atropine, hexamethonium and diphenhydramine, but not phentolamine, propranolol, and naloxone decreased the gallbladder pressure elevated by duodenal acidification. All these receptor blockers could not inhibit the effect of OP-CCK.
2. Block of the vagus by procaine could inhibit the effect of HCl. But vagal block and vagotomy at cervical trunk could not inhibit the effect of OP-CCK.
3. Previous administration of neostigmine facilitated the effect of HCl in subthreshold concentration... (More)

3. Previous administration of neostigmine facilitated the effect of HCl in subthreshold concentration but could not influence the effect of OP-CCK in subthreshold or superthreshold doses.
4. Lidocaine-HCl infused into the duodenum inhibited the effect of HCl while atropine-HCl first augmented and then inhibited the effect of HCl.
It is concluded that in the mechanism of gallbladder contraction induced by duodenal acidification, apart from the participation of CCK, there is a cholinergic nervous factor whose receptor is in the intestinal mucosa and whose action is to facilitate the release of CCK. Release of histamine may be involved in the mechanism. (Less)
