Regulation of Polyamine Analogue-Induced Apoptosis in Human Breast Cancer Cells

Abstract
Apoptosis is a defined process for cells to commit suicide through a set of biochemical and morphological cell changes. Activated apoptotic signals converge on the mitochondria, leading to permeabilization of mitochondrial membranes and release of different apoptosis regulatory proteins into the cytosol. With my studies I wanted to investigate the importance of polyamines in the regulation of apoptosis. Apoptosis is a defined process for cells to commit suicide through a set of biochemical and morphological cell changes. Activated apoptotic signals converge on the mitochondria, leading to permeabilization of mitochondrial membranes and release of different apoptosis regulatory proteins into the cytosol. With my studies I wanted to investigate the importance of polyamines in the regulation of apoptosis. The homeostasis of polyamines is highly regulated since polyamines are essential for cell proliferation and cell death.



A common feature of polyamine analogue treatment is inhibition of cell proliferation, but sometimes cells also die through apoptosis. When several different breast cancer cell lines were treated with polyamine analogues, mitochondrial activity increased indicating a cellular stress response. The stress response may be caused by polyamine analogue-induced polyamine depletion and/or by the presence of high concentration of a polyamine analogue that does not function as a natural polyamine. Polyamine analogue treatment of L56Br-C1 breast cancer cells resulted in a number of responses culminating in mitochondrial apoptotic cell death, as confirmed by the release of a number of apoptosis-related proteins from the mitochondria.



Another response to analogue treatment was the activation of the polyamine catabolic enzyme spermidine/spermine N1-acetyltransferase (SSAT). Using several different methods we have shown that there is an association of SSAT with the mitochondria implicating that SSAT and polyamine depletion are part of the apoptotic process induced by polyamine analogue treatment.



To investigate the role of the antiapoptotic protein Bcl-2 in polyamine analogue-induced apoptosis, a vector carrying the gene for Bcl-2 was introduced into the genome of L56Br-C1 cells. The degree of cell death, polyamine reduction, and SSAT activity decreased in Bcl-2 overexpressing cells compared to control cells.



In conclusion, polyamine analogue treatment clearly affects the mitochondria and can induce apoptosis as a response, something which can be prevented by Bcl-2 overexpression. (Less)

För att bibehålla balansen mellan olika typer av celler i kroppen finns flera olika mekanismer som reglerar celltillväxt och celldöd. Cancer uppstår när dessa reglermekanismer inte fungerar. Om behandlingen av kvinnor med bröstcancer ska kunna förbättras är det viktigt att man först studerar dessa mekanismer.



Generellt finns två olika former av celldöd, nämligen nekros och apoptos. Nekros uppkommer oftast i samband med en akut skada och resulterar i inflammation i den omgivande vävnaden, t.ex. när man får ett sår. Apoptos är cellens programmerade sätt att dö när den har en inre skada. Denna skada skulle kunna leda till att cellen blir farlig för hela individen, t.ex. när det har uppstått en mutation som kan orsaka cancer. Den apoptotiska celldöden är styrd via olika signalvägar som alla resulterar i en nedbrytning av cellen som gör att den dör utan att orsaka inflammation.



Det finns flera ämnen i celler som är nödvändiga för att balansen mellan celltillväxt och celldöd skall bibehållas. Inför min avhandling har jag studerat hur molekyler som heter polyaminer reglerar apoptosen via cellens energiproducerande organeller, s.k. mitokondrier. Polyaminer är naturligt förekommande molekyler som finns i bakterier, växtceller och djurceller. Det finns tre olika polyaminer, nämligen putrescin, spermidin och spermin. Då cellen är starkt beroende av att den inre polyaminhalten snabbt anpassas till dess behov måste cellen reglera polyaminnivån mycket noggrant. Detta gör cellen genom att ha en omsorgsfullt kontrollerad jämvikt mellan import, nytillverkning, nedbrytning och/eller export av polyaminer. Blir polyaminnivån för hög (giftigt) i cellen måste den bryta ner polyaminerna, medan allt för låga polyaminnivåer leder till en import eller nytillverkning. Till sin hjälp att reglera polyaminnivån har cellen olika enzymer. För tillverkning av polyaminer används enzymerna ornitindekarboxylas (ODC) och S-adenosylmetionindekarboxylas (AdoMetDC), medan nedbrytning av polyaminer sker med hjälp av enzymet spermidin/spermin N1-acetyltransferas (SSAT).



För att se hur polyaminerna påverkar och reglerar apoptos har jag behandlat olika bröstcancercellinjer med syntetiskt tillverkade substanser som liknar polyaminen spermin, s.k. sperminanaloger. Vid behandling med sperminanaloger tror cellen att den har för höga nivåer av spermin, vilket leder till att cellen startar nedbrytningen av polyaminer. Cellen aktiverar enzymet SSAT varpå polyaminerna bryts ner till mindre molekyler som lätt kan exporteras ut ur cellen. Samtidigt som SSAT aktiveras, minskar cellen aktiviteten för enzymerna ODC och AdoMetDC så att nybildningen av polyaminer avtar. Detta leder till att nivån av de naturligt förekommande polyaminerna kraftigt minskar och cellen följaktligen får polyaminbrist. När en omfattande polyaminbrist uppstår dör cellen via apoptos. När väl den apoptotiska processen har startats finns det ingen återvändo.



I mitt arbete har jag visat att sperminanaloger påverkar cellens mitokondrier. När man behandlar celler med polyaminanaloger tycks de bli stressade och då ökar aktiviteten i mitokondrierna. Vissa typer av bröstcancer celler dör cellen när de behandlas med sperminanaloger. När dessa celler dör genom apoptos är det inte bara de energiproducerande enheterna i mitokondrierna som påverkas utan även hela cellens utseende. Cellen får blåsliknande strukturer på cellytan och det bildas s.k. apoptotiska kroppar.



När jag undersökte var i cellerna SSAT fanns, visade det sig att det aktiva enzymet främst finns vid cellens mitokondrier. Mitokondrierna innehåller en mängd olika proteiner som hjälper till både att reglera och att finjustera den apoptotiska processen. När proteiner som framkallar apoptos frisätts från mitokondrierna ut i cellen startar en reaktionskaskad som slutar med apoptos. Skulle däremot proteiner som hämmar apoptosen frisättas startar aldrig reaktionskaskaden. Efter behandling med en sperminanalog frisläpps ett protein från cellens mitokondrier (cytockrom c) som kraftigt framkallar apoptos. När cytokrom c frisätts från mitokondrierna startar reaktionskaskaden omedelbart och cellen dör. Om cellen däremot har ett överuttryck av proteinet Bcl-2 i mitokondrierna, så minskar eller hämmas cytokrom c-frisättning vid behandling med en sperminanalog. Slutsatsen blir att balansen mellan proteiner som reglerar och finjusterar den apoptotiska processen bidrar till avgörandet om cellen ska överleva eller dö genom apoptos som framkallas genom behandling med en sperminanalog.



Sammanfattningsvis kan man säga att det är viktigt för cellen att inte bara noga reglera sina polyaminnivåer utan även att reglera balansen mellan olika mitokondriella proteiner. Efter behandling med olika sperminanaloger sjunker cellens polyaminnivåer kraftigt, varpå den apoptotiska processen startas. Denna process förstärks ytterligare med hjälp av apoptotiska proteiner som frisläpps från de stressade mitokondrierna. Dock kan ett överuttryck av Bcl-2 proteinet i mitokondrierna delvis eller helt skydda cellen mot apoptos trots att det råder polyaminbrist. Mina resultat tyder på att cellens sammansättning av en grupp mitokondriella apoptotiska proteiner skulle kunna vara en markör för en bröstcancers känslighet för polyaminanaloger. (Less)
