- Abstract
- Enzyme formulation development is a tedious process that requires lots of time and resources.
By improving parts of the formulation process, the formulation of enzymes could become more
efficient and allow for quicker commercialization of enzyme products. The focus of this project
has been on solid granular enzyme formulations, aiming to accelerate stability studies, develop
enzyme degradation prediction models and validate a small-scale screening system. The three
feed enzymes of phytase, amylase and protease were subjected to varying time, temperature,
pH and humidity to then be analyzed using mainly nanoDSF and activity assays. The results

of ingredients. Formulation development is a tedious process requiring lots of time, resources,
and money. Naturally, there are wishes to improve this procedure.
The three pillars to improve formulation development in this project are acceleration of
stability studies to obtain stability data faster, increase knowledge of enzyme degradation to
create empirical stability prediction models and lastly, preparation of small scale formulations
to enable higher throughput screening of formulation candidates or engineered protein
candidates.
The studied enzymes were phytase, protease and amylase in solid granular format due to its
common use in feed applications. These enzymes were subjected to varying conditions by
altering time, temperature, pH, and relative humidity. They were analyzed in regard to their
stability and degradation over time.
The results increased the general knowledge of enzyme degradation caused by the mentioned
parameters, providing the information of different degradation mechanisms at different
humidities and temperatures. The small-scale system proved successful and onwards, larger
stability screenings are possible. The results regarding acceleration of stability studies are
somewhat ambiguous and more data is still needed for clarity. (Less)
